Peptide Handling 101: Reconstitution, Storage, and Stability
A practical primer for research labs receiving lyophilized peptides for the first time.
Reconstitution and storage account for more reproducibility problems than almost any other variable in peptide research. This primer covers the basics.
Before you open the vial
Allow the sealed vial to reach room temperature before opening to avoid drawing moisture into the lyophilized cake.
Reconstitution
- Choose a diluent compatible with your assay (commonly bacteriostatic water).
- Inject slowly down the side of the vial — never directly onto the cake.
- Swirl gently; do not shake.
- Allow to fully dissolve before aliquoting.
Storage and stability
Lyophilized peptides are typically stable for 18–36 months at −20°C. Reconstituted solutions are typically used within 28 days when stored at 2–8°C.
